コンサルティング課題名：（例）～～～～に関わるタンパク質の単粒子解析に向けたクライオ電顕による性状評価
コンサルティング課題内容：
（持ち込む予定の複数のサンプルについて記述してください。均一で十分量のサンプルが精製出来ていることをアピールしてください。E0測定の支援期間は1年程度を目安としていただき、クライオ電顕単粒子解析に向けて、支援期間中に精製法の改良やグリッド作製条件の検討など、どこまで行うか具体的にお書きください。サンプルの形状が均一でない場合、他の因子を加えるなど形状を安定化させるための戦略があればそれもお書きいただければと思います。単粒子解析に適したサンプルが得られた場合は、そのサンプルに絞った申請を行うこともお書き添えください。）
（例）申請者のグループでは、これまで、XXX、YYY、ZZZについて、立体構造解析を目指した研究を行なってきた。しかし、これらについて、これまで数年に渡って結晶化を試みてきたものの、結晶を得ることができていない。均一かつ十分量のサンプルを精製することには成功しているので、近年、近原子分解能での立体構造解析が可能になった、クライオ電子顕微鏡による単粒子解析を試みたい。
サンプルの精製スキームの変更も含めて、クライオグリッド作製の条件検討やクロスリンクなどの条件検討を包括的に行っていきたいので、BINDS・解析領域の支援プログラム（E0測定）に申請する。また、XXX、YYY、ZZZの形状が不安定な場合、これらの因子のみでは単粒子解析に適したサンプルを得られない可能性もある。そのため、因子の形状を安定化させうる相互作用因子との複合体についても、クライオ電子顕微鏡での解析対象に加える。
以上を扱っていく過程で、単粒子解析に適したサンプルが見つかった場合は、再度、そのサンプルに焦点を絞ったE1測定の申請を行いたい。上記の計画の成否に関わらず、支援期間は1年で終了とする。その期間を過ぎた場合、再申請を行うか、全体計画の見直しを行いたい。

Cryo-EM-based characterization of [example]-related proteins for single-particle analysis
Consulting Project Content:
Please describe the multiple samples you plan to bring. Emphasize that you have purified a sufficient amount of uniform samples. Assuming a support period of approximately one year for E0 measurements, please specify concretely how far you will go in improving purification methods and examining grid preparation conditions during the support period to prepare for cryo-EM single-particle analysis. If the sample shape is not uniform, please describe any strategies to stabilize the shape by adding other factors. If a sample suitable for single-particle analysis is obtained, please also write that you will submit an application focusing on that sample.
[Example] Our group has been conducting research aimed at determining the three-dimensional structures of XXX, YYY, and ZZZ. Although we have attempted crystallization for several years, we have not obtained crystals. Since we have succeeded in purifying sufficient amounts of homogeneous samples, we would like to try single-particle analysis using cryo-electron microscopy, which has recently enabled near-atomic resolution structure determination.
We will apply for the BINDS analysis domain support program (E0 measurement) to comprehensively examine cryo-grid preparation conditions, cross-linking conditions, and even modifications to the sample purification scheme. Additionally, if the shapes of XXX, YYY, and ZZZ are unstable, we may not be able to obtain samples suitable for single-particle analysis using these factors alone. Therefore, we will also include complexes with interaction factors that can stabilize the shapes of these factors as targets for cryo-EM analysis.
If we identify a sample suitable for single-particle analysis during this process, we plan to submit another application for E1 measurement focused on that sample. Regardless of the success of this plan, the support period will end in one year. After that period, we will either reapply or review the overall plan.
